ABSTRACT

As a traditional Chinese medicinal material, Citri Sarcodactylis Fructus is often used as medicine after steaming to reduce its pungency and give full play to its medicinal effects. By consulting the relevant literature, this paper intends to sort out the related researches on the processing history, modern processing standard records, flavor and meridian tropism, efficacy, processing and taking methods, processing technology, chemical composition, pharmacological effects and quality analysis of Citri Sarcodactylis Fructus. It is found that the processing methods of Citri Sarcodactylis Fructus in the past dynasties are mainly roasting, distilled into dew, aging, frying, salting and steaming, of which steaming is the most common. In modern times, steaming is mainly used in Guangdong and Sichuan. The descriptions of flavor and meridian tropism and efficacy of Citri Sarcodactylis Fructus in the past dynasties are basically consistent with the modern descriptions. Its taste is pungent, bitter and acidic, and it is warm in nature. It belongs to the liver, spleen, stomach and lung meridians. Citri Sarcodactylis Fructus contains volatile oil, flavonoids and their glycosides, coumarins and limonins and other compounds. In recent years, high performance liquid chromatography (HPLC), gas chromatography-mass spectrometry (GC-MS) and ultra performance liquid chromatography-mass spectrometry (UPLC-MS) and other methods are commonly used for the chemical composition analysis. Citri Sarcodactylis Fructus has the activities of relieving cough, reducing phlegm, relieving asthma, anti-inflammation, anti-bacteria and anti-oxidation, etc. After processing, its volatile and non-volatile components, and pharmacological effects all have a certain change. However, the current research on processing of Citri Sarcodactylis Fructus is not in-depth enough. It is necessary to further analyze the material basis of steaming to reduce dryness, explore its dryness medicinal substances and dryness-effect quantitative correlation, so as to clarify its processing mechanism, and provide basis for the subsequent processing research, resource development and comprehensive application of Citri Sarcodactylis Fructus in the future.

ABSTRACT

ObjectiveTo reveal the medication rules of the clinical prescriptions containing Citri Sarcodactylis Fructus, and to provide a basis for the modern clinical application of Citri Sarcodactylis Fructus, the development of health products and the research and development of new drugs. MethodThe clinical prescriptions containing Citri Sarcodactylis Fructus were retrieved from CNKI, Wanfang Data, and VIP, and then a basic database of prescriptions was established via the traditional Chinese medicine inheritance auxiliary platform (V2.5), IBM SPSS V20, and Excel (Microsoft Office 2016). The frequency and association rules of the medicines in prescriptions (compatible medicines) and the corresponding syndromes were then mined. ResultThe prescriptions were screened according to the inclusion and exclusion criteria. A total of 458 clinical prescriptions containing Citri Sarcodactylis Fructus were collected, involving 388 Chinese medicines, and the total frequency of medicines reached 6 199. The core compatible medicines (frequency > 130) of Citri Sarcodactylis Fructus included Poria (frequency of 222), Atractylodis Macrocephalae Rhizoma (217), Paeoniae Radix Alba (196), Bupleuri Radix (159), and Citri Reticulatae Pericarpium (142). The Citri Sarcodactylis Fructus-compatible medicines with frequency > 49 were selected for further analysis, which included 34 medicines with the cumulative frequency of 3 131 (50.51% of the total frequency). These medicines mainly have the functions of tonifying Qi, invigorating Qi, tonifying blood, alleviating edema and promoting urination, promoting digestion, and activating blood and relieving pain. They are mainly warm, cold, or mild-natured, taste bitter, sweet, or acrid, and have the tropism in the spleen, liver, stomach, or lung meridians. The association rule analysis demonstrated that 14 medicine combinations were commonly used, and the core combinations were Poria-Citri Sarcodactylis Fructus, Atractylodis Macrocephalae Rhizoma-Citri Sarcodactylis Fructus, Paeoniae Radix Alba-Citri Sarcodactylis Fructus, Bupleuri Radix-Citri Sarcodactylis Fructus, Atractylodis Macrocephalae Rhizoma-Poria-Citri Sarcodactylis Fructus, and Citri Reticulatae Pericarpium-Citri Sarcodactylis Fructus. The clinical prescriptions containing Citri Sarcodactylis Fructus were mainly used to treat 52 diseases corresponding to 11 types of traditional Chinese medicine (TCM) syndromes. Three representative syndrome types, including spleen and stomach syndromes, Qi-blood-body fluid syndromes, and gynecological syndromes were selected for further association rule analysis. In the treatment of spleen and stomach syndromes, the core compatible drugs were Atractylodis Macrocephalae Rhizoma, Poria, Paeoniae Radix Alba, Bupleuri Radix, Citri Reticulatae Pericarpium, and Pinelliae Ehizoma, which, together with Citri Sarcodactylis Fructus, formed 25 commonly used medicine combinations (16 combinations composed of 2 medicines and 9 combinations composed of 3 medicines). In the treatment of Qi-blood-body fluid syndromes, the core compatible drugs were Poria, Paeoniae Radix Alba, Atractylodis Macrocephalae Rhizoma, Astragali Radix, Hordei Fructus Germinatus, and Bupleuri Radix, which, together with Citri Sarcodactylis Fructus, formed 23 common medicine combinations (17 combinations composed of 2 medicines, 5 combinations composed of 3 medicines, and 1 combination composed of 4 medicines). In the treatment of gynecological syndromes, the core compatible medicines were Atractylodis Macrocephalae Rhizoma, Paeoniae Radix Alba, Angelicae Sinensis Radix, Astragali Radix, Cyperi Rhizoma, and Poria, which constituted 25 common medicine combinations (15 combinations composed of 2 medicines and 10 combinations composed of 3 medicines). ConclusionWe employed the traditional Chinese medicine(TCM) inheritance auxiliary platform to explore the compatibility of Citri Sarcodactylis Fructus-containing clinical prescriptions and the corresponding TCM syndromes, which intuitively showcased the medication rules of Citri Sarcodactylis Fructus. Specifically, Citri Sarcodactylis Fructus was mainly combined with the medicines for tonifying Qi, invigorating Qi, tonifying blood, alleviating edema and promoting urination, promoting digestion, and activating blood and relieving pain to treat different TCM syndromes. While soothing liver, regulating Qi, harmonizing stomach, and relieving pain, the combinations tonify and activate blood, invigorate spleen, and resolve dampness. The findings are of great significance to the rational application of Citri Sarcodactylis Fructus, the development of health food, and the research of new drugs and will bolster the development of Chinese medicine industry.

ABSTRACT

Objective:To establish the high performance liquid chromatography (HPLC) fingerprint of Citri Sarcodactylis Fructus, and to search for makers to characterize the quality difference of Citri Sarcodactylis Fructus from different origins coupled with chemometrics. Method:The analysis was performed on a Thermo Hypersil GOLD C<sub>18</sub> column (4.6 mm×250 mm, 5 μm) with mobile phase consisted of acetonitrile-0.05% phosphoric acid solution for gradient elution, and the detection wavelength was set at 254 nm. A total of 31 batches of samples were analyzed to establish the HPLC fingerprint of Citri Sarcodactylis Fructus. Similarity evaluation was performed by Traditional Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System (2012 edition) to confirm the common peaks, which were identified by comparison of reference substances. On the basis, chemometrics methods were used to analyze and evaluate the quality of Citri Sarcodactylis Fructus from different origins. At the same time, 3 batches of 5 species of decoction pieces from the genus <italic>Citrus</italic> in the family Rutaceae, including Citri Sarcodactylis Fructus, Aurantii Fructus Immaturus, Aurantii Fructus, Citri Reticulatae Pericarpium Viride and Citri Reticulatae Pericarpium, were randomly collected for evaluating the effectiveness and reliability of the established HPLC fingerprint of Citri Sarcodactylis Fructus. Result:HPLC fingerprint of Citri Sarcodactylis Fructus was established and 22 common peaks were identified. And seven common peaks among them were identified as 6,7-dimethoxycoumarin, diosmin, hesperidin, byakangelicin, 5,7-dimethoxycoumarin, bergapten and oxypeucedanin. Except for 2 batches of samples, the similarities of fingerprints between other 29 batches of samples were >0.9. The 31 batches of Citri Sarcodactylis Fructus were basically divided into 3 groups by cluster analysis and principal component analysis, which were consistent with the classification of three different producing areas. Eight differential markers were screened by orthogonal partial least squares discriminant analysis and four of them (5,7-dimethoxycoumarin, bergapten, 6,7-dimethoxycoumarin and diosmin) were identified by reference substances. Similarity evaluation of 5 species of decoction pieces from genus <italic>Citrus</italic> in the family Rutaceae was carried out by taking the reference fingerprint of Citri Sarcodactylis Fructus as treference chromatogram, similarity of Citri Sarcodactylis Fructus decoction pieces was 0.892-0.977, and the similarities of the other 4 kinds of decoction pieces were 0.215-0.517. Conclusion:The established fingerprint method is reasonable, effective and accurate for quality control of Citri Sarcodactylis Fructus, the characterization information is more comprehensive combined with chemometrics.

ABSTRACT

Citri Sarcodactylis Fructus is a both medicinal and edible species specified by the China Ministry of Health, with a long history in China. According to the ancient monographs about materia medica, it was found that the records of the Citri Sarcodactylis Fructus on the original plants were confused. This paper reviewed the ancient monographs about materia medica, and made a summarization and textual research on the name, origin, habitat, processing methods, medicinal properties and clinicacy efficacy of Citri Sarcodactylis Fructus based the comprehensive analysis on modern literatures and authoritative books of Chinese herbal medicine. The results indicated that there were many bynames of Citri Sarcodactylis Fructus. Before the Yuan Dynasty, there was a mixed use of Citri Sarcodactylis Fructus and Citri Fructus, which were not distinguished from each other in terms of nature and taste until the Yuan dynasty. Citri Sarcodactylis Fructus was a varietas of Citri Fructus. The main shape of the original plant of Citri Sarcodactylis Fructus is "like a human hand with fingers" as recorded in ancient monographs about materia medica. The main places of origin of Citri Sarcodactylis Fructus were Guangdong, Zhejiang, Fujian, Sichuan, which were relatively stable. There were fewer records about medicinal proces-sing methods of Citri Sarcodactylis Fructus. Only steaming and baking methods were found in ancient monographs about materia medica, and the steaming method could reduce the irritability of Citri Sarcodactylis Fructus. The processing of therapeutic dietary of Citri Sarcodactylis Fructus was widely used in folk, which was represented by Chaozhou Laoxianghuang, a traditional succade made of Citri Sarcodactylis Fructus. According to the 2015 edition of Chinese Pharmacopoeia, Citri Sarcodactylis Fructus had effects in soothing liver and regulating gas, relieving pain in the stomach, eliminating dampness and resolving phlegm, which was basically consistent with the descriptions in ancient monographs about materia medica. This paper defined the original plant of Citri Sarcodactylis Fructus, and sorted out and summarized the processing methods, nature and taste of Citri Sarcodactylis Fructus, so as to provide data support for the standardization of the processing technology and the development and utilization of Citri Sarcodactylis Fructus.

ABSTRACT

Objective::To analyze and identify the chemical constituents of Citri Sarcodactylis Fructus by an ultra-high performance liquid chromatography coupled with hybrid quadrupole-orbitrap high-resolution mass spectrometry (UPLC-Q-Orbirap HRMS) method. Method::Compounds were separated on Thermo Scientific Accucore™ C18 column (3 mm×100 mm, 2.6 μm). The mobile phase was 0.1% formic acid solution and 0.1% formic acetonitrile solution. The flow rate was 0.3 mL·min-1, and the column temperature was set at 30 ℃. HRMS was performed using an electrospray ion source (ESI), and scanned in a positive ion mode by means of full scan/data dependent secondary scan (Full MS/dd-MS2). Compound Discoverer 3.0 software that could be linked to mzCloud network database and local high-resolution mass spectrometry database of traditional Chinese medicine components was used to analyze the data, based on accurate molecular mass, retention behaviors and characteristic ion fragmentation of the compounds, as well as literature information and relevant reference materials. Result::Totally 54 chemical constituents in Citri Sarcodactylis Fructus were identified, including 16 flavonoids, 17 coumarins, 3 limonoids, 6 nucleosides and nucleobases, 2 organic acids, 3 aromatic aldehydes, 1 amino acid and 6 other components. Conclusion::The established UPLC-Q-Orbitrap HRMS method can be used to effectively and rapidly identify main chemical constituents of Citri Sarcodactylis Fructus. The findings provide a methodological reference and theoretical foundation for defining the pharmacodynamic material base and optimizing quality control index of Citri Sarcodactylis Fructus, which is of guiding significance for further development and utilization of the resources.

ABSTRACT

Citri Sarcodactylis Fructus, a traditional Chinese medicine with dual-purpose of drug and food, has a long history of cultivation and application, showing high medicinal, dietetic, ornamental and economic values. Many modern researches have been conducted on the extraction technology, chemical constituents, quality standards and biological activities, but the herbal textual researches were relatively insufficient. The related records of Citri Sarcodactylis Fructus in the ancient Chinese medicinal classics were ambiguous and confusing. By referring to ancient Chinese medicinal classics and modern books, the textual researches of Citri Sarcodactylis Fructus, including the names, plant morphology, origin, medicinal properties, efficiency and application were systematically reviewed and analyzed in this paper, in order to provide the basis for the confirmation of original source of Citri Sarcodactylis Fructus. The results showed that Citri Sarcodactylis Fructus was usually recorded as ‘Xiangyuan’ ‘Juyuan’ ‘Ganyuan’ ‘Xiangyuan’ ‘Foshougan’ etc. in ancient books. After the Ming and Qing dynasties, it became clear that ‘Xiangyuan’ was equivalent to ‘Juyuan’, but different from ‘Foshougan’. Therefore, these two species were recorded separately. From the ancient times to the present day, Citri Sarcodactylis Fructus was always mainly distributed in south of the Yangtze River, such as Fujian, Guangdong, Guangxi, and Jiangxi provinces. Ancient records and modern reports basically have the same understanding on properties, efficacies and indications of Citri Sarcodactylis Fructus, considering it was warm in nature with the effectiveness of soothing liver, relieving pain, eliminating dampness and resolving phlegm. ‘Baking it into powder on new tiles’ and ‘steaming’ were the traditional processing methods. The description of ‘The longer of storage period, the better of the effect’ was occasionally recorded. These results provide theoretical references for the further study, resource development and comprehensive application of Citri Sarcodactylis Fructus.

ABSTRACT

Objective:To compare volatile components in raw products and steamed products of Citri Sarcodactylis Fructus. Method:Volatile oil in different batch of raw products and steamed products of Citri Sarcodactylis Fructus was extracted by steam distillation, and volatile components were identified by GC-MS.The determination was performed with injector temperature of 280℃, high pure helium(≥ 99.999%) as carrier gas, split ratio of 10:1, and sample size of 3 μL with temperature programming.Mass spectrum conditions included electron bombardment ion source, electron bombardment energy of 70 eV, ion source temperature of 230℃, acceleration voltage of 34.6 V, resolution of 2 500, scanning range of m/z 40-350.Peak matching, cutting and noise filtering were used in analyzing data based on GC-MS combined with orthogonal partial least square-discriminant analysis(OPLS-DA). Result:A total of 58 components were separated and identified from the different batches of raw Citri Sarcodactylis Fructus and their processed products, and 27 differential chemical components were identified by multivariate statistical analysis, among them, 15 common differential components presented different changing laws.The relative contents of α-pinene, β-pinene, p-cymene, β-linalool, L-4-terpinenol, α-terpineol, nerol, β-cyclocitral, geraniol and α-citral in raw products were higher than that of their corresponding processed products.The relative contents of β-caryophyllene, cis-α-bergamotene, γ-muurolene, γ-elemene and β-bisabolene in processed products were higher than that of their corresponding raw products. Conclusion:The content and composition of volatile components in Citri Sarcodactylis Fructus from Guangdong province have changed after being steamed.The results can provide experimental basis for expounding the processing principle and quality evaluation of Citri Sarcodactylis Fructus, and have positive significance for promoting the research and development of this Lingnan characteristic decoction pieces.

ABSTRACT

This study was aimed to establish fingerprints of Citri Sarcodactylis Fructus from different geographical origins,and to use the method of pattern recognition to compare the differences of Citri Sarcodactylis Fructus from different habitats.In this study,high performance liquid chromatography (HPLC) was used to establish fingerprints for 25 batches of Citri Sarcodactylis Fructus from 4 habitats.Furthermore,similarity evaluation,cluster analysis (CA) and principal component analysis (PCA) were performed.The results from established fingerprints showed that a total of 26 common peaks were pointed out and 4 peaks were identified as the common peaks.The CA and PCA can be used to compare Citri Sarcodactylis Fructus from different habitats.It was concluded that Cirri Sarcodactylis Fructus in near geographic origins had a higher similarity,while the different geographic origins had a higher difference.

ABSTRACT

Bergamot essential oil with the fragrance of flowers in pure and fresh smell is a kind of volatile oil extracted from fresh Citri Sarcodactylis Fructus. Like orange and lemon, it is a kind of natural perfume oil. Bergamot essential oil, with obvious antibacterial and anti-inflammatory effects, can treat sunburn, psoriasis, acne, improve skin, etc. It also can create a relaxed and pleasant feeling and thereby has the effect of antidepressant. Bergamot essential oil has been considered as rare spice oil, and has high value of application in the food and cosmetics industry in foreign countries. At present, the researches on bergamot essential oil by domestic and foreign scholars mainly focus on the optimization of extraction process, component analysis, pharmacological action, and other aspects. With the continuous developments and applications of bergamot essential oil and related products, the range of study for bergamot essential oil is getting more and more widely. This paper reviews the researches on the extraction technologies, various components analysis, pharmacological effects, and applications of bergamot essential oil. At the same time, all kinds of problems probably occurred in the process of bergamot essential oil researches are discussed and the corresponding solutions are pointed out. Finally, some possible development trends and the prospects of the research on bergamot essential oil are also related. Thus more information about the research progress of various aspects of bergamot essential oil can be obtained and serve the research and product development of bergamot essential oil better in our country.
